I am getting low coupling efficiency with amidites, any suggestions?
There are two factors that can affect the coupling efficiency, amount of time and dryness of the acetonitrile. It may be helpful to increase the coupling time and if you believe the quality of the acetonitrile may have been compromised use molecular sieves to make sure it is anhydrous.
